ABSTRACT

Objective To investigate the entry point, direction and fixation range of Magic screw for acetabular posterior column fracture. Methods The 3D models of 100 pelvis were reconstructed by Mimics 19. 0 software based on CT data, and the virtual magic screw were placed in acetabular posterior column, then the screw' s entry point were determined, and the direction, length, diameter and safe range were measured respectively. Acetabular posterior column with magic screw was osteotomy modeling by Mimics 19.0 and imported the Geomagic Wrap 2017 software, after that anatomical parameters of the screw path were measured. Results The entry point of Magic screw on bone surface was located at (33. 37±5. 53) mm of the anterior inferior iliac spine moving posteriorly, and ( 13. 40±3. 70) mm of the apex of the upper margin of acetabular moving celphalad in male, which were (33. 97±5. 46) mm and (9. 01±3. 86)mm in female. The posterior inclination angle, interior inclination angle of magic screw and the angle between screw and iliac wing were (57.40±6.57)°, (52. 09±5. 65)° and (15. 21 ±3.42)° in male, which were (55. 64±8. 01)°, ( 51. 55±5. 58)° and (9. 85±3. 68)°in female. The maximum diameter of male screw was (6. 97±0. 98) mm, which was (6. 39±0. 85) mm in female. The length of male screw was (76. 73±9. 20) mm, which was (63. 64±8. 37) mm in female. The safe range of posterior inclination and interior inclination of were ( 7. 19 ± 3. 30)° and (9. 41 ±3. 95)° in male screw with 5.5 mm diameter, which were (8. 37±2. 82)° and ( 10. 32±3. 93)° in female screw with 5. 2 mm diameter. In the direction of the screw, the length range of the posterior column which is fixed by screw was (56. 87±7. 60) mm, and the proportion of fractures fixed with screws which were located on the top of the acetabular was 20/50 in male, which were (41. 71±7. 97) mm and 8/50 in female. Conclusion Percutaneous Magic screw is a minimally invasive treatment for acetabular posterior column fracture, which is difficult to operate. The screw can fix the fracture which is located at the middle and upper part of the acetabular posterior column.

ABSTRACT

Objective research the effects of metformin on proliferation and apoptosis in nasopharyngeal carcinoma cell CNE-1 and investigate the role of miR-let-7a、IGF-1R in it. Methods The nasopharyngeal carcinoma cell CNE-1 was treated with different concentrations of metformin for 24h, then the proliferation activity of cell was detected by CCK8 method; the apoptosis rate of cell was measured by flow cytometry; the expression levels of bcl-2、bax and IGF-1R mRNA and miR-let-7a were detected by real-time quantitative PCR; the expression level of IGF-1R protein was detected by Western blot. Results Compared with the control group, the cell proliferation activity of metformin group decreased(P<0.05), and it gradually decreased along with the increase of metformin concentration. The cell apoptosis rate of metformin group increased(P<0.05 except for the 5 mmol/L group), and it gradually increased along with the increase of metformin concentration. The expression levels of bax mRNA and miR-let-7a were up-regulated in the metformin group(P<0.05), while the expression levels of bcl-2 mRNA,IGF-1R mRNA and IGF-1R protein were decreased(P<0.05). Conclusion Metformin could inhibit the proliferation and induce apoptosis of CNE-1. The mechanism maybe related to the up-regulation of miR-let-7a and down-regulation of IGF-1R.

ABSTRACT

<p><b>BACKGROUND</b>Diabetic nephropathy is a major cause of renal failure in diabetes mellitus (DM). It has been known that renin-angiotensin system (RAS) blockers have a renal protective effect. This study aimed to investigate whether treatment with angiotensin II receptor blocker, olmesartan, could modify renal hemodynamic variables and vascular structural properties, then attenuate renal injury in streptozotocin (STZ)-induced DM rats.</p><p><b>METHODS</b>DM was induced in male Wistar rats by intraperitoneal administration of STZ. The rats were then randomized to a DM group and an olmesartan treatment (OLM + DM) group. The normal group (non-DM) were administered only citrate buffer. At the end of the 14th week, blood glucose, kidney weight/body weight and urinary protein-to-creatinine ratio were determined. Further, the flow-pressure and pressure-glomerular filtration rate (GFR) relationships were determined for maximally vasodilated, perfused kidneys. From the relationship, 3 indices of vascular structural properties were estimated: slope of flow-pressure (minimal renal vascular resistance, reflecting overall luminal dimensions of preglomerular and postglomerular vasculature), slope of pressure-GFR (glomerular filtration capacity against pressure) and threshold pressure for beginning filtration at pressure-GFR (preglomerular to postglomerular vascular resistance ratio). Kidneys were then perfusion fixed for histological analysis. The renal histopathology was observed by light microscopy.</p><p><b>RESULTS</b>The body weight of DM rats was lower than that of non-DM rats. Blood glucose, kidney weight/body weight, urinary protein-to-creatinine ratio were significantly greater in DM rats than in non-DM rats. The parameters such as kidney weight/body weight, urinary protein-to-creatinine ratio in OLM + DM rats had dramatically decreased compared with those in DM rats. However, the treatment with olmesartan had no effect on blood glucose levels. The slope of flow-pressure relationship was greater in DM rats than that in non-DM rats (P < 0.05). But the slope of the pressure-GFR relationship was lower in DM rats than that in non-DM rats (P < 0.05) with the x-intercept of the line similar between the two groups. The slope of the flow-pressure relationship was decreased in DM rats group treated with olmesartan (P < 0.05). Moreover, olmesartan significantly increased the slope of the pressure-GFR relationship in DM rats (P < 0.05). The x-intercept of the pressure-GFR relationship reduced following olmesartan in DM rats.</p><p><b>CONCLUSIONS</b>Treatment with olmesartan reduced urinary protein-to-creatinine ratio independent of blood glucose and increased average renal vessel lumen diameter in the perfused kidneys of STZ-induced DM rats, predominantly in preglomerular vessels, and then improved renal excretory capability. These findings were consistent with remodeling of the preglomerular vasculature in our hisological measurements.</p>

ABSTRACT

<p><b>BACKGROUND</b>Rapamycin (RAPA) is a relatively new immunosuppressant drug that functions as a serine/threonine kinase inhibitor to prevent rejection in organ transplantation. RAPA blocks activation of T-effector (Teff) cells by inhibiting the response to interleukin-2. Recently, RAPA was also shown to selectively expand the T-regulator (Treg) cell population. To date, no studies have examined the mechanism by which RAPA converts Teff cells to Treg cells.</p><p><b>METHODS</b>Peripheral CD4(+)CD25(-) naive T cells were cultivated with RAPA and B cells as antigen-presenting cells (APCs) in vitro. CD4(+)CD25(-) T cells were harvested after 6 days and analyzed for expression of forkhead box protein 3 (Foxp3) using flow cytometry. CD4(+)CD25(+)CD127(-) subsets as the converted Tregs were isolated from the mixed lymphocyte reactions (MLR) with CD127 negative selection, followed by CD4 and CD25 positive selection using microbeads and magnetic separation column (MSC). Moreover, mRNA was extracted from converted Tregs and C57BL/6 naive CD4(+)CD25(+) T cells and Foxp3 levels were examined by quantitative real-time polymerase chain reaction (rt-PCR). A total of 1 x 10(5) carboxyfluorescein succinimidyl ester (CFSE)-labeled naive CD4(+)CD25(-) T cells/well from C57BL/6 mice were cocultured with DBA/2 or C3H maturation of dendritic cells (mDCs) (0.25 x 10(5)/well) in 96-well round-bottom plates for 6 days. Then 1 x 10(5) or 0.25 x 10(5) converted Treg cells were added to every well as regulatory cells. Cells were harvested after 6 days of culture and analyzed for proliferation of CFSE-labeled naive CD4(+)CD25(-) T cells using flow cytometry. Data were analyzed using CellQuest software.</p><p><b>RESULTS</b>We found that RAPA can convert peripheral CD4(+)CD25(-) naive T Cells to CD4(+)Foxp3(+) Treg cells using B cells as APCs, and this subtype of Treg can potently suppress Teff proliferation and maintain antigenic specificity.</p><p><b>CONCLUSION</b>Our findings provide evidence that RAPA induces Treg cell conversion from Teff cells and uncovers an additional mechanism for tolerance induction by RAPA.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of rapamycin in inducing naïve murine effector T cell (Teff) convert to regulatory T cell (Treg) in vitro.</p><p><b>METHODS</b>The forkhead box protein 3 (FoxP3) negative Teff were isolated and purified from the spleen and lymph node of C57 BL/6 murines aged 6-8 weeks, then Teff were cultured in three groups with mature dendritic cells (mDC), B cells, and plate coated Anti-CD3. In addition, the control wells and the test wells were prepared in each group, rapamycin were not added in the control wells but added in the test wells with concentrations of 1, 10, 50, and 100 nmol/L. Percentages of FoxP3 positive Treg were examined by flow cytometry after 4 days in Anti-CD3 group and after 6 days in the other two groups.</p><p><b>RESULTS</b>As shown by the flow cytometry, the percentages of FoxP3 positive Treg were as follows in three group: in the mDC group, it was 0.01% in the control well and 0.39%, 0.47%, 0.34%, and 0.26% in test wells; in B cell group, it was 0.01% in the control wells and 5.56%, 5.89%, 7.15%, and 4.72% in the test wells; in Anti-CD3 group, it was 0.93% in the control wells and 1.35%, 1.07%, 1.02%, and 1.19% in test wells. No significant difference was found between the test wells and control wells in the mDC group and Anti-CD3 group; however, the percentages of FoxP3 positive Treg was significantly different between the test wells and control wells in the B cell group (P < 0.01).</p><p><b>CONCLUSION</b>When B cell is acted as the antigen-presenting cell, rapamycin can effectively induce Teff convert to Treg in vitro.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the relationship between aging and the number and function of bone marrow-derived endothelial progenitor cells (EPC) in rats.</p><p><b>METHODS</b>The number and function of bone marrow-derived EPC were measured in 4, 10 and 18 months old rats (n = 10 in each group). Mononuclear cells (MNC) were isolated from rat bone marrow by Ficoll density gradient centrifugation and cultured for 7 days, EPC were identified as adherent cells double positive stained for FITC-UEA-I and DiI-acLDL under laser confocal immunofluence microscopy. Boyden chamber was used to determine the migration capacity of EPC and the number of recultured EPC was counted to test the adherent function of EPC.</p><p><b>RESULT</b>The number (42.0 +/- 5.8 vs 30.8 +/- 4.7 vs 21.5 +/- 4.7 per field) and the migration and adhesion capacities of EPC were significantly reduced with aging.</p><p><b>CONCLUSION</b>This study shows that the number and function of EPC are decreased with aging.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To observe the characteristics of uterine contraction and stages of labor during delivery under continuous epidural block anesthesia.</p><p><b>METHODS</b>Totaling 213 parturients in spontaneous labor under epidural block anesthesia with dilated cervical orifice of 3 cm were monitored for the contraction cycle, duration, intensity and curve types of uterine contraction, and recordings were made for 30 min before and 30, 60 and 120 min after the anesthesia took effect, respectively. The duration of the active phase in the first, second and third stages of labor was compared between 421 cases with anesthesia and 237 without anesthesia.</p><p><b>RESULTS</b>Significant difference was noted in the objective indexes of uterine contraction recorded after anesthesia had taken effect (P<0.05) in comparison with those before anesthesia, suggesting significantly attenuated uterine contraction after anesthesia, whereas these indexes underwent no significant further variation as compared between different time points after anesthesia (P>0.05). The average active phase in the first stage was significantly shorter in anesthesia group than that in the control group (P<0.05), but the average duration of the second and third stages of labor differed little between the two groups with appropriate use of oxytocin under strict monitoring (P>0.05). The rates of obstetric forceps utilization and use of oxytocin were higher in anesthesia group than in the control group (P<0.05).</p><p><b>CONCLUSION</b>Epidural block anesthesia produces certain influences on uterine contraction and stages of labor during delivery, for which appropriate treatment measures may prove beneficial.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the relationship of TCM Syndromes, involving Viscera Syndrome and involving Meridian Syndrome, with blood hypercoagulative state and insulin resistance in patients with diabetic ischemic stroke.</p><p><b>METHODS</b>Insulin sensitivity index (ISI) could reflect the insulin resistance, and those reflecting blood hypercoagulative state parameters such as platelet agglutination test (PAgT), fibrinogen (FG), in vitro thrombus length (VTL) and activating partial thrombinogen time (APTT), were used to expound and prove the relationship with the stroke syndrome.</p><p><b>RESULTS</b>The symptom scores, PAgT, VTL, FG levels in patients involved with Viscera Syndromes (including those of Wind Syndrome, Phlegm-Dampness Syndrome, Heat-Fire Syndrome and Yin-deficiency with Yang-excess Syndrome) were significantly higher than those in patients involved with Meridian Syndromes (P < 0.05 or P < 0.01), while the ISI level in the former was lower than that in the latter (P < 0.01).</p><p><b>CONCLUSION</b>There is close relationship between blood hypercoagulative state, insulin resistance and TCM Stroke Syndromes in patients with diabetic ischemic stroke.</p>
